Fungal DNA Detection Kit (PCR-Fluorescent Probe Method) [HYM005]
Product Code: HYM005
Specification: 50 tests/kit
Detection Limit:5 CFU/mL
Product Overview
The Fungal DNA Detection Kit (PCR-Fluorescence Probe Method) is used in conjunction with the Fungal and Bacterial DNA Extraction and Purification Kit (Magnetic Bead Method) to qualitatively detect the presence of fungal contamination in cells, cell products, vaccines, and other products.
This kit utilizes fluorescent probe qPCR technology, with primers and probes designed for conserved fungal gene fragments. Based on sequence alignment results, it allows for the qualitative detection of thousands of fungal species. The assay demonstrates high specificity, with no cross-reactivity observed for mycoplasma species closely related to fungi (e.g., Mycoplasma orale, Mycoplasma hyorhinis, Mycoplasma pneumoniae) or common engineered cells such as CHO, VERO, and HEK293. Validated in accordance with relevant regulatory requirements, the limit of detection reaches 5 CFU/mL, complying with pharmacopoeial sensitivity requirements for NAT methods as an alternative to culture methods.
Product Performance
SpecificityMatrix Effect: Test sample matrices (basal medium, fetal bovine serum) showed no interference with the detection results.Cross-reactivity: No cross-reactivity occurs with bacteria (Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureus, Clostridium perfringens) or engineered cells (Vero cells, HEK293 cells, 293T cells, CHO-K1 cells)
Limit of Detection (LOD)
| Reagent kit nameStrain Name | Strain CFU/mL | Positive Rate(Positive/Total) |

| Aspergillus niger | 3 | 100%(24/24) |
Stability
| Stability Study Projects | Details |
| Freeze-Thaw Stability | The kit was tested after 3 and 5 repeated freeze-thaw cycles respectively, with an RSD of < 5%. |
| Accelerated Stability | Two different batches of the kit were used to test the same sample simultaneously. The results were consistent, with consistent Ct values for the positive control and internal control showing no significant differences. All were successfully detected, with an RSD of < 5%. |
| Inter-batch Stability | The kit was tested on different instrument models including ABI 7500, Bio-Rad CFX96, Bioer FQD-96A, and Roche LightCycler 480II, and all yielded valid detection results. |
